4D blood flow mapping using SPIM-microPIV in the developing zebrafish heart

چکیده
Fluid-structure interaction in the developing heart is an active area of research in developmental biology. However, investigation of heart dynamics is mostly limited to computational fluid dynamics simulations using heart wall structure information only, or single plane blood flow information so there is a need for 3D + time resolved data to fully understand cardiac function. We present an imaging platform combining selective plane illumination microscopy (SPIM) with micro particle image velocimetry (μPIV) to enable 3D-resolved flow mapping in a microscopic environment, free from many of the sources of error and bias present in traditional epifluorescence-based μPIV systems. By using our new system in conjunction with optical heart beat synchronisation, we demonstrte the ability obtain non-invasive 3D + time resolved blood flow measurements in the heart of a living zebrafish embryo.

4-Dimensional light-sheet microscopy to elucidate shear stress modulation of cardiac trabeculation.
Hemodynamic shear forces are intimately linked with cardiac development, during which trabeculae form a network of branching outgrowths from the myocardium. Mutations that alter Notch signaling also result in trabeculation defects. Here, we assessed whether shear stress modulates trabeculation to influence contractile function. Specifically, we acquired 4D (3D + time) images with light sheets b...

Mitral valve regurgitation (MVR) is a common valvular disease in dogs. Hydrokinetic evaluation of the blood flow within the ventricles has become possible by vector flow mapping (VFM), which shows the blood flow within the ventricles in vector and vortex flows. Blood flow within the left ventricle of MVR dogs was compared at different stages of MVR and to that of normal dogs. 14 normal dogs and...
متن کامل3D Visualization of Developmental Toxicity of 2,4,6-Trinitrotoluene in Zebrafish Embryogenesis Using Light-Sheet Microscopy
Environmental contamination by trinitrotoluene is of global concern due to its widespread use in military ordnance and commercial explosives. Despite known long-term persistence in groundwater and soil, the toxicological profile of trinitrotoluene and other explosive wastes have not been systematically measured using in vivo biological assays. Zebrafish embryos are ideal model vertebrates for h...
متن کاملComprehensive 4D velocity mapping of the heart and great vessels by cardiovascular magnetic resonance
BACKGROUND Phase contrast cardiovascular magnetic resonance (CMR) is able to measure all three directional components of the velocities of blood flow relative to the three spatial dimensions and the time course of the heart cycle. In this article, methods used for the acquisition, visualization, and quantification of such datasets are reviewed and illustrated. METHODS Currently, the acquisiti...
متن کاملIn Vivo Wall Shear Measurements within the Developing Zebrafish Heart
Physical forces can influence the embryonic development of many tissues. Within the cardiovascular system shear forces resulting from blood flow are known to be one of the regulatory signals that shape the developing heart. A key challenge in investigating the role of shear forces in cardiac development is the ability to obtain shear force measurements in vivo. Utilising the zebrafish model sys...
